[Abstract]

Objective: To study the effects of simvastatin on human prostate cancer cell line PC-3 in vitro and its mechanisms. Meanwhile, to investigate the effects of simvastatin combined with anti-cancer agents Paclitaxel?Cisplatin on human prostate cancer cell line PC-3 in vitro.Methods: 1 PC-3 cells were incubated in culture medium in vitro. Effect of simvastatin on the proliferation of PC-3 cells was measured by MTT colorimetric method and the OD values were computed. Apoptosis and distribution of cell cycle were examined with flow cytometry. The morphological changes of PC-3 cells were observed by light microscopy and the analysis of DNA-fragment was shown by DNA gelatum electrophoresis. In addition, expression of p21Ras?E2F-1?CDK4 and Bax was analyzed by flow cytometric indirect immunofluorescent technigue. 2 MTT colorimetric method was performed to evaluate the potential cytostatic effect of combining simvastatin and Paclitaxel?Cisplatin on human prostate cancer cell line PC-3 and the OD values were computed.
